Prime probably isn't going to help so I wouldn't worry about adding any of it. Water changes and finding the source are your only options; you might consider looking at the source water for ammonia as well.

Have you noticed any corals dying or fish missing? How do things look in the aquarium? This will be an indication of a real problem or faulty water test.

A couple of fish that you had in the tank turned up missing a few weeks ago?

What kind of ammonia problem are we talking about, what does the test kit read and what brand of test kit are you using? If there was a high level of ammonia in the water, you would see an immediate response by your corals.

Prime will only treat the symptom, not the cause.... I would put an APB out for the missing fish!!! (sorry couldn't resist the pun). Keep up the water changes and maybe add some sort of "live bacteria" like Cycle or whatever that claims to help cycle the tank in the beginning. Maybe your bacteria bed needs a kick in the pants!!
