Should I prime and paint the pieces before I assemble or after it is screwed together? I would think that priming and painting first then touching up after would help seal it better. Is this correct?

It really isn't necessary. Wood glues are meant to be used on bare wood, you need to fill any joint gaps with wood filler, and the final joint seal will be from layers of two part epoxy paint and a bead of silicone after the box is complete. There might even be adhesion problems with the glue and filler putty.

Thanks Desert Fish. I will paint it after it is put together. Oh, I changed the height from 4 feet to 3 feet. Bad enough the 1/2" glass panel is over $200. I couldn't deal with the price of the 1" panel.
